Full-marker reagent portfolio supports mechanistic studies linking Hippo, TGF-β/BMP and WNT signalling cascades
HUSTON, TX, UNITED STATES, October 20, 2026 /EINPresswire.com/ — The Hippo signalling pathway does not operate in isolation. It forms intricate crosstalk networks with two canonical cascades: Wnt and TGF-β/BMP. This crosstalk represents a major research hotspot, and unravelling these interactive mechanisms is critical for understanding the pathogenesis of tumours, bone metabolic disorders and fibrotic conditions.
To begin with the Hippo-TGF-β/BMP axis: upon ligand stimulation, TGF-β and BMP activate cell-surface receptors, triggering SMAD1/2/3/4 transcription factors to translocate into the nucleus and regulate target-gene expression. YAP/TAZ directly binds SMAD proteins to synergistically amplify transcription of pro-fibrotic and pro-osteogenic genes. Hippo pathway inactivation leads to massive nuclear accumulation of YAP, which exaggerates TGF-β-driven tissue fibrosis. By contrast, high LATS kinase activity disrupts YAP-SMAD complex formation and suppresses pathological bone overgrowth as well as organ fibrosis. Concurrent dysregulation of both pathways serves as a core pathogenic feature in osteoporosis, osteoarthritis, hepatic and pulmonary fibrosis.
For Hippo-WNT crosstalk: WNT activation triggers nuclear translocation of β-catenin to initiate proliferative gene transcription. YAP forms a co-transcriptional complex with β-catenin to synergistically boost tumour cell proliferation. Polarity proteins CDH1 and CTNNA1 participate in upstream regulation for both pathways. Disruption of cell-cell junctions simultaneously activates WNT signalling and dampens Hippo activity, jointly fuelling tumour invasion and metastasis. Such dual-pathway crosstalk is essential evidence for studies covering embryonic development, stem-cell proliferation and intestinal neoplasms.
(Figure 1 Core biological functions and central mechanism of the Hippo signalling pathway. The Hippo pathway governs cell proliferation and apoptosis to regulate organ size and maintain cell-number homeostasis. It mediates cell contact inhibition by sensing cell density and restraining excessive proliferation. It modulates bone metabolism and remodelling by controlling osteogenesis, osteoclastogenesis and bone repair. Aberrant YAP/TAZ activity promotes tumour-related proliferation and invasion. It cooperates with TGF-β to mediate fibrosis and matrix deposition. Core mechanism: MST1/2 → LATS1/2 → phosphorylation and nucleocytoplasmic shuttling of YAP/TAZ.)
Investigating multi‑pathway crosstalk requires simultaneous measurement of core Hippo components, TGF‑β/BMP members, and upstream/downstream WNT markers. This calls for broad target coverage and multi‑assay compatibility. Poor target selection or technique‑limited antibodies can disrupt workflows and produce incomplete mechanistic data. Cloud‑Clone’s products cover 97 targets at key nodes of these interconnected pathways to support mechanistic studies of multi‑pathway crosstalk.
(Figure 2 List of Hippo pathway-related targets from Cloud-Clone, including core catalogue numbers for each biomarker.)
This product suite includes core Hippo molecules such as YAP, TAZ, MST and LATS; complete TGF-β/BMP targets including TGFB1/2/3, SMAD1-4 and BMP2/4/6/7; as well as full WNT regulatory proteins: WNT1/3A/4/11, CTNNB1, AXIN1/2 and the complete FZD family. All antibodies have undergone practical multi-platform validation, supporting quantitative Western blot on cell lysates, immunohistochemical localisation in tissue sections, immunofluorescence co-localisation and flow-cytometry-based protein quantification.
For co-localisation assays, Cloud-Clone AF488- and FITC-conjugated secondary antibodies pair with pathway-specific primary antibodies. Researchers can label YAP and β-catenin within the same cell to directly visualise their nuclear co-localisation and validate pathway synergy. Flow cytometry enables parallel quantification of multiple pathway proteins and is compatible with Luminex and CBA multiplex assays, permitting high-throughput profiling of numerous targets from limited sample input.
Cloud-Clone antibodies are validated for human, mouse, rat, porcine, bovine and other model organisms, suiting human clinical samples, gene-edited mouse models and large-animal translational research. Monoclonal antibodies deliver high specificity for flow cytometry and quantitative Western blot applications, while polyclonal antibodies offer strong affinity for staining low-abundance proteins in tissue samples. Matching recombinant proteins and ELISA kits are also available, enabling end-to-end workflows from protein qualitative localisation to precise sample quantification.
As an original reagent manufacturer performing full in-house production, Cloud-Clone provides complete traceable validation datasets for all products to reduce experimental trial-and-error costs. This comprehensive biomarker portfolio enables researchers to build solid, logically consistent regulatory models and generate reliable experimental evidence for multi-pathway interaction projects.
